Doubt if its the diff lock making the noise,the diff lock is spring loaded to disengage and it is external,its mounted outside the brake on the right side,take the weight off the wheels and check the bearings for slop,the most likely thing to be causing a clunk every revolution is the ring gear or one of the two reduction gears in the transaxle,the best thing for you to do is drain the oil and remove the plate from the back of the transaxle and have a look and see what is wrong,the 580C axle is not conventional i.e the halfshaft to not come out to the wheels from the centre of the diff like a auto or truck,the two shafts that do come out from the diff centre carry the brakes on each side,those shafts are in mesh with two large reduction gears that fit on the inner end of the halfshafts,if one tooth has come off one of those gears it will cause a clunk,or if the main ring gear it self has stripped a tooth,so take the cover off and have a look before the related part gets chewed up.
